About

I am Dr. K. S. Dinesh Babu, currently working as an Assistant Professor (Regular/Permanent faculty) in the Department of Botany and Microbiology, Hemvati Nandan Bahuguna Garhwal University (A Central university), Tehri Garhwal, Uttarakhand - 249161. Previously, I was working as a senior project associate in CSIR-Center for Cellular and Molecular Biology, Hyderabad, India. I have completed my Ph. D (Biotechnology) degree in 2019 from the Plant Biotechnology Department, Madurai Kamaraj University, with the title “Functional investigation on Bhendi yellow vein mosaic virus proteins in plant pathogenesis”. I am specialized in functional genomics of plant and plant viral genes and have 5 years of working experience with Arabidopsis thaliana and Nicotiana benthamiana transgenic plant generation, confirmation of transgene integration, and protein localization via fluorescent microscopy.

My Ph. D research work involves the study of host pathogen interactions between Bhendi yellow vein mosaic virus (BYVMV) and its permissible host N. benthamiana. In the preliminary experiments, I have identified the interference of BYVMV on the host mediated transcriptional gene silencing via Chop-PCR and bisulfite-next generation sequencing methods. Through site directed mutagenesis, I have created two separate BYVMV mutant constructs containing stop codons in the N-terminal of C2 and C4 genes and analyzed the role of these viral genes in transcriptional gene silencing (TGS) suppression. Through Bisulfite-NGS sequencing method, I have identified the level of TGS suppression by both mutants and wild type infectious BYVMV. Through viral DNA quantification (qPCR) methods, I have identified the indispensable role of BYVMV C4 protein in viral movement and involvement of C4 protein in TGS suppression. I have analyzed the effect of mutation on viral DNA replication by protoplast transfection and the viral quantity determination method. I have identified the BYVMV C4 protein as a dispensable factor for the replication of BYVMV viral DNA. This part of the work has been published in MDPI Journal “Epigenome” with title “DNA methylation suppression by Bhendi yellow vein mosaic virus”. I did extensive bioinformatics analysis on C-terminal acidic transcription activation domain of begomoviral AC2/C2 proteins and its variable amino acid composition.

I am well trained in Geminiviral infectious clone preparation, agroinfiltration, circular viral DNA isolation and quantification by qPCR (Quantitative PCR), viral protein localization in plant cells using viral protein-Fluorescent protein (GFP) fusion constructs, transgenic plant production via Agrobacterium mediated gene transfer, evaluation and confirmation of transgene integration in transgenic plants via genomic DNA PCR, Southern hybridization, reverse transcription PCR and immuno detection of target proteins. I have practiced Bioinformatic analysis of DNA, protein sequences, motifs and having strong working knowledge of light, fluorescence and confocal microscopy. I am well trained in plant internal morphology (Plant Anatomy) analysis and have three years of research experience on the culture of plant organs under various chemically defined media.

My scientific interests include plant functional genomics, transgenic plant research and plant molecular virology research. Best of my abilities include, learning new research techniques and applying the technology to current laboratory research, quickly accommodating to the new research projects, helping my coworkers in all day to day research and data analysis, taking care of all types of general laboratory works, having a good command of the English language, designing and performing scientific research and interpretation of results are my key strengths. I will communicate my work progress regularly and can also assist on writing project reports. I have the ability to handle multiple tasks carefully while working on different projects.
