Research Focus: "Isolation and Characterization of Novel Lytic Bacteriophages Against Enterococcus faecium and Its Clinical Isolates"
Research on phage therapy and its potential applications in treating infections such as urinary tract infections, bacteremia, and biofilm disruption or inhibition using bacteriophages. Collaborating with the Institute of Medical Sciences, Banaras Hindu University. I have Submitted two Enterococcus faecium phages to the NCBI database:
RG1 Phage (Accession ID: PQ586903, December 10, 2024)
SSBHU Phage (Accession ID: PQ778307, February 4, 2025).
My Doctoral research focused on understanding how housekeeping (SigA) and alternative sigma factors (SigB) regulate transcription and bacterial adaptation in adverse environmental condition in Mycobacterium smegmatis. I performed in silico characterization of sigma factor classes across Mycobacteria where I found interesting outcome, which includes motif, domain and conserver amino acid residue followed by cloning, expression, and purification of SigA and SigB. Using biophysical analysis, I demonstrated distinct stability patterns due to different secondary structural composition of peptides alpha helical and beta pelted sheet. Finally, through affinity purification–HRMS proteomics, I mapped the interactomes of SigA and SigB during log and stationary phases, identifying key regulators that involve in the regulation of metabolic pathways they. This work provides important insights into transcriptional regulation, stress response, and bacterial survival mechanisms.
