Dr. Saubhik Som completed his Ph.D. with Prof. Sandeep Eswarappa in the Dept of Biochemistry, Indian Institute of Science, Bengaluru, India. His PhD project focused on the physiological significance of stop codon readthrough in a mitochondrial membrane protein MTCH2. He studied a MTCH2 readthrough-deficient 3′UTR knockout transgenic mouse and showed that MTCH2 readthrough is important in maintaining MTCH2 protein levels, and the deletion results in reduced skeletal muscle ATP levels, oxidative phosphorylation and musculoskeletal activity in mice, due to altered mitochondrial translation. This work supports the hypothesis that stop codon readthrough has evolved as a biologically meaningful mechanism that can quickly fine-tune protein levels and cellular physiology in a tissue-dependent manner (Som et al., manuscript under preparation).
During his PhD, he also developed a rapid in vivo assay that uses subcutaneous injection of plasmid DNA into the mouse tail to study post-transcriptional gene regulation and can be used for in vivo screening of therapeutics (Som et al., FEBS Letters, 2025).
