University Agra, he worked on the ‘Design and development of carbohydrate-containing molecules as chemotherapeutic agents at Banaras Hindu University, Varanasi, India (with Dr. V. K. Tiwari as his mentor), starting in 2012 and obtained his Ph.D. degree in 2016. He was offered a research associate position at Indian Institute of Technology, Patna, India (2016-2017), before starting his independent research career at IGNTU, he worked on the activation of glycosyl donors and their application in O-glycosylation reaction as a National Postdoctoral Fellow at the Indian Institute of Technology, BHU, India (2017-2019), and subsequently on the Ruthenium catalyzed olefinic coupling and their application in natural product synthesis as Institute post-doctoral fellow at Indian Institute of technology, Kanpur, India (with Prof. D. H. Dethe as his mentor) in (2019-2021). He has contributed as corporate researcher and worked as senior research scientist at Jubilant Biosys, Noida, India (2021-2022) and at NSJ Prayog Life Sciences, Tarnaka, Hyderabad, India (2022-2023). During his research carrear, he has awarded with different national fellowship schemes, (NPDF awarded by DST-SERB India-2017; Research associate awarded by CSIR-2016, India; Junior Research fellowship-2012 was awarded by UGC, India; IPDF awarded by IIT-Kanpur-2019; PDF awarded by Free State University-2019, South Africa).
Our research is mainly focused in the area of Organic/Carbohydrate/Medicinal chemistry. Our previous detailed synthetic research work was involved for the click reaction in carbohydrate chemistry, Staudinger reaction for sugar derivatives, functionalization of carbohydrate with oxozolidine-2-thione and thiozolodine-2-thione, BCF prompted glycosylation reaction for O-glycoside synthesis and Ruthenium prompted olefinic coupling and application in natural product synthesis etc.
Previously, we have contributed a lot for the synthesis of medicinal valued novel glycol-hybrid molecules; synthesis of sugar based morpholine fused triazoles and their analogs have been developed via two steps process (i) one pot methodology involving multi-component Cu (I) catalyze click reaction of glycosyl alkynes, NaN3 and epichlorohydrin in aqueous medium and second step gave morpholine fused 1,5-triazoles using one-pot reaction of azido alcohols with propargyl bromide followed by subsequent intra-molecular cyclo-addition reaction. We have developed different libraries of glycoconjugates of ethisterone, noscapine, quercetin, vanilen etc with various sugar azides using copper catalyzed clicke reaction. The glycoconjugates were investigated for their antiliesmanial, anti-cancer activities. Further, we studied the catalytic activity of highly efficient and reusable pre-formed mono- and dinuclear heteroleptic copper(I) dithiocarbamate and dithiocarbimate complex for CuAAC and developed diverse triazolyl vaniline glycoconjugate. Next, we explored the triazole chemistry and established metal free different catalytic systems for [3 + 2] cycloaddition reaction of glycosyl azides with nitro-olefins to give 1,5-disubstituted 1,2,3-triazolyl glyco-conjugates. A competitive study between p-TsOH and diverse ammonium salts including phase transfer catalysts and ionic liquids have been discussed to search the best one for glyco-conjugation.
Apart, triazole chemistry, we have also attempted for the synthesis of oxazolodin-2-thione and thiozolodin-2-thone via treating to diverse azidoalcohols with, carbondisulfide, TPP in toluene in heating condition. Reaction proceeds through the Staudinger reaction to yield desired .sugar based oxa- and thiozolodin-2-thione derivatives. Same chemistry has been applied to develop carbohydrate based thiourea. Various azides were heated with amines in presence of CS2 and TPP in toluene. During the course of my first postdoctoral research work, herein, I have also worked in the area of carbohydrate chemistry. Initially, tris(pentafluorophenyl)borane promoted stereoselective glycosylation with trichloroacetimidate glycosyl donors and glycals were explored. The reactions proceed efficiently with a wide range of acceptors, from sugar to non-sugar, under mild conditions in the presence of a catalytic amount of B(C6F5)3.. A practical method for the selective and controlled oxidation of thioglycosides to corresponding glycosyl sulfoxides and sulfones is reported using urea–hydrogen peroxide (UHP). During the course of my second post-doctoral research, I have explored my chemistry in the area of natural product synthesis. A Ru-catalyzed direct oxidative coupling reaction of vinyl ketones was developed. It offers a straightforward and atom-economical protocol for synthesis of functionalized (E,E)-1,6-dioxo-2,4-diene derivatives.
