About
Gaetano Montelione is an internationally recognized expert and innovator in the fields of protein NMR and structural genomics. He is Endowed Constellation Chair in Structural Bioinformatics at Rensselaer Polytechnic Institute, and Distinguished Professor Emeritus and inaugural Jerome and Lorraine Aresty Chair in Cancer Biology, Rutgers University.
Montelione carried out pioneering work on NMR pulse sequence development, including the design of the first triple-resonance protein NMR experiments with G. Wagner, as well as subsequent development of HCCcoNH-TOCSY and other widely-used NMR experiments. This research led Montelione to develop software using artificial intelligence for automated analysis of protein resonance assignments and structures. The resulting technology platform provided the basis for many key contributions by NMR to structural genomics and de novo protein design. Montelione has also made key contributions to elucidating the structural basis by which influenza viruses suppress host innate immune response, and in structure-function studies of oncogenes and tumor suppressors.
Montelione's current efforts focus on NMR studies of larger, more challenging systems, including protein complexes and integral membrane proteins. He leads the structural biology components of several biomedical collaborations, including projects on BET proteins associated with cancer biology, protein design, and viral-host protein/RNA interactions. Recognizing new opportunities provided by recent advances in applying artificial intelligence (AI) to protein structure modeling, over the last few years the lab has focused on the scientific goal of modeling alternative conformational states of proteins by combining AI prediction methods with experimental data. For example, they have developed the AlphaFold-NMR (AISAR) approach for modeling multiple conformational states of proteins and applied this to enzymes and protein complexes. Montelione currently serves as advisor to the wwPDB, the Biological Magnetic Resonance Database, and to the Critical Assessment of Protein Structure Prediction (CASP). Montelione also leads the monthly CASP Special Interest Group webinar on "Modeling Conformational Ensembles of Biological Macromolecules".
