My long term research goal is to investigate mechanisms of cardiac remodeling in diabetes and metabolic syndrome, and to identify viable targets for cardiac regeneration and prevention of cardiac arrhythmia. We have described a novel, intracellular component of the renin-angiotensin system (RAS), called intracrine RAS, in the heart and its significance in diabetic cardiomyopathy. Additionally, our new data has pointed towards the role of the intracellular RAS in the regulation of cardiac conductance, thus arrhythmias, through novel protein: protein interactions with XIRP1. Intracellular Ang II also modulates nuclear localization of proteins that control proliferation of cardiomyocytes. We are studying the regulation, role, and mechanism of the intracrine RAS in the heart using in vitro and in vivo approaches. We utilize knockout and transgenic mice in combination with pharmacological models of diabetes and metabolic syndrome. We perform in vitro cell signaling studies using neonatal and adult cardiomyocytes. We have expertise in echocardiography and catheter-based hemodynamic measurements to study heart function. Other approaches used in my laboratory include mass spectrometry, real-time RT-PCR, Western analysis, histology, and confocal microscopy.
