About
My PhD and post PhD both work was in the field of apatmer. The initial period of work gave me opportunity to understand aptamer, how it can be selected and way of using it in detecting the target. Later in post PhD work, I extended my work on same field and target with new approaches. In post PhD, I was able to understand, how selection of aptamers can be done in quick time as conventional method takes lots of time and labor intensive. Use of Non SELEX method of aptamer selection drastically reduces time. Further different strategies were used to detect target. I used ALISA in conventional 96 well plate as well as with magnetic beads. This was a new approach as beads are not meant for this purpose. We carried out experiment in real sample to get information of aptamer detection capabilities. Further modification in aptamers makes them more resistant to any attack from nuclease action. In dot blot assay different types of membrane was used to optimize detection method. In detection method itself several strategies were adopted to get best possible result. We also learn that by evaluation of G score (QGSR Mapper) and analysis of complex structure of aptamers (M- fold software) is sufficient to select synthetic DNA for biosensor development insisted for going for dissociation constant determination. These all information gave me confidence in using aptamers in other ongoing projects under my PI. I selected aptmers against specific targets in semen sexing and pregnancy detection project of cattle. Aptamers were successfully generated in these project and showed good detecting result. Apart from research work, I was also involved in IPR related project. In post PhD dork at NBAIM, I learnt how to draft an application for patent filing. I was also involved in copyright filing. The PhD and post PhD work gives me ample knowledge of bench work as well as information on commercialization of product.
